(AP) ? President Barack Obama is addressing his most solidly supportive constituency, black voters, at the end of a four-day, five-state trip on Wednesday.

After a fundraising swing through the Seattle and Portland, Ore., Obama arrived in New Orleans to address the National Urban League, a civil rights group.

While enthusiasm among some Obama supporters has faded since 2008, support for America's first black president remains high among African-Americans. In a recent Associated Press-GfK poll, 82 percent of black adults said they would vote for Obama. His approval rating among blacks was 87 percent.

Obama skipped an appearance at the NAACP earlier this month, raising questions about whether his campaign was taking black voters for granted. The White House blamed a scheduling conflict and sent Vice President Joe Biden to address the nation's oldest civil rights organization instead.

Republican challenger Mitt Romney also addressed the National Association for the Advancement of Colored People, raising eyebrows when he told the crowd: "If you want a president who will make things better in the African-American community, you are looking at him."

The audience booed the presumptive Republican nominee when he said he would repeal "Obamacare" ? the name critics use for Obama's health care overhaul ? if elected.

In his speech to the National Urban League, Obama was to announce that he will sign an executive order on Thursday that creates a new office to bolster the education of African-American students. The White House says the office will coordinate the work of communities and federal agencies to ensure that these youngsters are better prepared for high school, college and career.

Obama also was raising campaign money at two separate stops in New Orleans: an intimate 20-person event at the cost of $25,000 per person, and a larger event at the House of Blues for 400 people, with tickets starting at $250.

Obama raised at least $6 million at events this week in California, Oregon and Washington. The president started the trip Sunday with a stop in Colorado to meet with survivors and families of the victims of last week's movie theater massacre.

Coal miner Peabody 2Q earnings slip

ST. LOUIS (AP) ? Peabody Energy's second-quarter profit fell with so many utilities using cheaper natural gas in place of coal.

The world's biggest private-sector coal company trimmed its 2012 production outlook and forecast lower-than-expected earnings for the third quarter. Its stock price slid, though the latest results beat Wall Street expectations.

The coal industry and its stocks have been battered ? St. Louis-based Peabody's shares dropped 16 percent during the second quarter ? as utilities switched to cheap natural gas from coal to generate electricity. Natural gas prices are the lowest in years because of huge supplies from booming production. And the mild winter across much of the nation didn't put much of a dent in the gas surplus.

Natural gas prices did hit a high for the year Tuesday as scorching temperatures sent people scampering for the air conditioner, but prices are still well below last year, and a quarter of what they were just four years ago.

Peabody's net income totaled $204.7 million, or 75 cents per share, in the second quarter, down from $284.8 million, or $1.05 cents, a year earlier. Revenue totaled $2.0 million, on par with $1.98 billion a year earlier. Wall Street expected Peabody to earn 53 cents per share on revenue of $2.06 billion, according to FactSet.

Citing what he called "a choppy market environment," Peabody chairman and chief executive Gregory Boyce said "the industry experienced significant headwinds in the second quarter, with declining global thermal coal prices as well as low U.S. natural gas prices driving record high U.S. utility coal stockpiles."

Still, he said, second-quarter imports by China ? the world's second-biggest economy and a key Peabody market ? hit record levels, and third quarter global prices for coal used in making steel have settled at higher levels. Natural gas prices in the U.S. also have rebounded more than 50 percent from their springtime lows, Peabody said.

"Peabody is weathering the macroeconomic storms well," Boyce told analysts during a conference call. Yet "Peabody is guarded in our near-term view of global market fundamentals."

"U.S. coal markets have shown some positive signals, but significant recover is not yet at hand," Boyce added, believing U.S. coal use will rebound next year as natural gas prices rise. "We continue to be cautious, given European recession, China's deceleration and high stockpiles that persist in the U.S."

The company forecast third-quarter earnings at 20 to 45 cents per share, well below the 65 cents a share estimated by analysts surveyed by FactSet. Peabody also pared its 2012 coal sales expectations by 5 million tons from its April projection, to between 230 million and 250 million tons.

Shares of Peabody fell $2.30, or 9.9 percent, to $20.86 in midday trading Tuesday.

Peabody's earnings are closely watched because the company usually is among the first of the coal sector's big players to report each quarter, giving analysts a snapshot of the industry's health. Peabody rival Arch Coal Inc., which last month said it would lay off about 750 workers in the Kentucky, Virginia and West Virginia coalfields, reports its second-quarter earnings Friday.

"We had thought a positive result from (Peabody) could have sprung a trap on the bears' big short positions in the sector, but this result won't do the job," JPMorgan analyst John Bridges wrote in a research note Tuesday to clients. "Other coal companies with a more U.S.-driven bottom line could report more positively and lift the group, but the cautious tone in Peabody's commentary is unlikely to lift the cloud that hangs over this sector."

Peabody said that industry data last month showed that coal again was the fastest-growing major fuel last year, rising 5.4 percent as the only fossil fuel to record above-average growth. Coal now accounts for 30.3 percent of global energy consumption, the highest share since 1969.

In the U.S., Peabody has mines in Illinois, Indiana, Wyoming, Arizona and New Mexico. It also owns mines in Australia.

AMD game console chip exec departs for NVIDIA - Neowin

The executive at AMD chiefly responsible for getting the company's graphics chips inside the Xbox 360 and the Wii game consoles has now left the company. The Wall Street Journal reported today that Bob Feldstein departed the company on Friday and came to work for one of its biggest rivals, NVIDIA, this week.

The story states that Feldstein previously worked at ATI before AMD acquired the company in 2006. By then, ATI had already signed deals with Microsoft and Nintendo to bring a version of its Radeon graphics chips to the Xbox 360 and Wii game consoles, respectively.

Feldstein, who had the official title of corporate vice president of business development while at AMD also reportedly has signed a deal with Nintendo to put a Radeon-based chip inside the upcoming Nintendo Wii U console. The story adds that he also secured similar deals for the next game console from Sony and Microsoft, even though neither company has commented publicly on their next-gen game console plans.

His new role at NVIDIA will be as its vice president of technology licensing. You can bet that NVIDIA, which got its GeForce chip inside the Playstation 3, would like to have its hardware inside future game consoles as well.

In a move that promises to bring the advantages of computer aided design (CAD) to genetic engineers, the first computer model of a complete bacterium has been produced in the US. It means researchers will soon be able to modify models of an organism's genome on a computer screen - or create artificial lifeforms - without the risks of undertaking wet biology in secure biosafety labs.

The pathogen is called Mycoplasma genitalium, a bacterium implicated in a number of urethral and vaginal infections. The bug was ripe for modelling say researchers at Stanford University in California, because it has the smallest genome of any free-living organism, with just 525 genes. By contrast, the popular lab pathogen E. coli has 4288 genes.

The modelling was undertaken by bioengineer Markus Covert and colleagues. To get the raw data for their model, they undertook an exhaustive literature review - spanning 900 research papers - to allow them to program into their model some 1900 experimentally observed behaviours and molecular interactions that M. genitalium can take part in during its life cycle.

In software terms, they found the behaviours of the 525 genes could be described by 28 algorithms, each governing the behaviour of a software module modelling a different biological process. "These modules then communicated with each other after every time step, making for a unified whole that closely matched M. genitalium's real-world behaviour," claims the Stanford team in a statement. Their research appears in the journal Cell (doi: 10.1016/j.cell.2012.05.044).

Such models will ultimately give biologists the freedom to undertake "what if" scenarios common in regular engineering - changing parameters in a genome design, say, like a civil engineer adjusts the width of a bridge deck on a computer to see what happens. As well as being experimentally useful, allowing artificial organisms and synthetic lifeforms to be created virtually (harming no-one), they could also boost biosafety by preventing accidental creations of lethal pathogens. In 2001, for instance, researchers in Australia accidentally created a lethal strain of mousepox.

In a commentary article in Cell, systems biologists Peter Freddolino and Saeed Tavazoie of Columbia University say they hope the work will soon be extended to more commonly used lab bugs like E. coli - but also warn that the technique's accuracy has yet to be demonstrated. It is unclear, they say, "how well overall behaviors will be predicted from a collection of separately obtained parameters" gleaned from hundreds of research papers.

But the US National Institutes of Health, which funded the modelling work, is excited. It believes the model a major step towards finding "new approaches for the diagnosis and treatment of disease", says James Anderson, an NIH program director.

Genetic mutations that cause common childhood brain tumors identified

ScienceDaily (July 22, 2012) ? Researchers at the Stanford University School of Medicine and Lucile Packard Children's Hospital have identified several gene mutations responsible for the most common childhood brain tumor, called medulloblastoma, adding evidence to the theory that the diagnosis is a group of genetically distinct cancers with different prognoses. These and accompanying findings are likely to lead to less-toxic, better-targeted treatment approaches over the next two years, the researchers said.

"We tend to treat all medulloblastomas as one disease without taking into account how heterogeneous the tumors are at the molecular level," said Yoon-Jae Cho, MD, an assistant professor of neurology and neurological sciences at Stanford, a pediatric neurologist at Packard Children's and the senior author of the new research. "This paper represents a finer-grained view of the genetic landscape of these tumors and provides us with some leads on how to develop new therapies."

The research, which appeared online in Nature July 22, is part of a large, ongoing effort to characterize genetic errors in medulloblastoma. Two companion studies on which Cho is a co-author will be published simultaneously with his paper. The three papers came from a consortium that involves scientists at Stanford, Packard Children's, the Broad Institute, Children's Hospital Boston, the Dana-Farber Cancer Institute, the German Cancer Research Center, Brandeis University and the Hospital for Sick Children in Toronto.

Current treatment for medulloblastoma, which originates in the cerebellum and affects about 250 U.S. children each year, begins with surgery to remove as much of the tumor as possible. Patients then receive a combination of radiation and chemotherapy, but the treatments are not tailored to the tumor's genetic characteristics.

Cho's team extracted DNA from 92 medulloblastoma tumors and compared it with DNA from matched blood samples from the same patients, uncovering 12 significant "point mutations" -- single-letter errors in the genetic code -- that occurred frequently in the brain cancer. A handful of the mutations had been previously identified in smaller studies of medulloblastoma, but several mutations were novel in both medulloblastoma and in cancer.

Among the newly identified mutations was one in an RNA helicase gene, DDX3X, which Cho said is the second-most common mutation in medulloblastoma tumors. "Mutations in this gene have now also been identified in other tumor types, such as chronic lymphocytic leukemia, and head and neck tumors," he said.

However, the researchers found that it was rare for the same gene mutated in several different patients' tumors. More commonly, mutations involving a set of genes regulating a single biological pathway were found in the tumors -- a pattern that is emerging across cancer genome sequencing efforts.

Though no single tumor in the study carried all 12 mutations, the researchers were able to categorize the tumors according to which mutations they possessed. "We now understand that there are certain tumors with particular genetic signatures that are really resistant to standard treatments," Cho said. Children with medulloblastoma do not routinely have their tumors' genetic signatures characterized, but Cho believes that such characterization coupled with targeted therapies could greatly enhance tumor treatment.

About two-thirds of medulloblastoma patients now survive five years past diagnosis, but many survivors suffer lasting physical or intellectual side effects from their cancer treatments. Drugs tailored to a tumor's genetic profile have the potential to save more patients while reducing side effects, Cho said.

Several of the mutations discovered affect cellular signals that switch large groups of genes on and off. "The dysregulation of these 'epigenetic programs' is becoming a common theme not only in medulloblastoma but across cancer," Cho said. Such pathways may be good targets for cancer drugs; indeed, drugs targeting one such pathway (histone methyltransferases) are currently in pre-clinical development, while agents against another pathway (Hedgehog signaling pathway) are entering phase-2 clinical trials for medulloblastoma.

Cho is the co-chair of a committee within the Pediatric Brain Tumor Consortium that guides which drugs should be moved into clinical trials next. "Our plan is that within the next one to two years we will be able to offer kids a new set of compounds that have a clear biological rationale based on our genomic studies." Cho said. "We want to make sure we're being careful of what we move forward with, but at the same time, for some of these kids we don't have many, if any, effective and durable treatment options."

Cho's collaborators at Stanford included research associate Furong Yu; Gerald Crabtree, PhD, professor of pathology and of developmental biology and a member of the Stanford Cancer Institute; and life science research assistant Amanda Kautzman.

The research was funded by the National Institutes of Health, a St. Baldrick's Foundation Career Development Award, the Beirne Faculty Scholar endowment at Stanford University, German Cancer Aid, the BMBF ICGC-PedBrain project, the Howard Hughes Medical Institute, the Pediatric Brain Tumor Foundation, the Canadian Institutes of Health Research, the Hospital for Sick Children and the Mullarkey Research Fund. Cho consults for Novartis to help develop biomarkers for the company's clinical trial design.
